Neurite development in PC12 cells cultured on nanopillars and nanopores with sizes comparable with filopodia

چکیده
We investigated the effect ofnanoscale topography on neurite development in pheochromocytoma (PC12 cells) by culturing the cells on substrates having nanoscale pillars and pores with sizes comparable with filipodia. We found that cells on nanopillars and nanopores developed fewer and shorter neurites than cells on smooth substrates, and that cells on nanopores developed more and longer neurites than cells on nanopillars. These results suggest that PC12 cells were spatially aware of the difference in the nanoscale structures of the underlying substrates and responded differently in their neurite extension. This finding points to the possibility of using nanoscale topographic features to control neurite development in neurons.

Effects of Different Concentrations of Morphine on Staurosporine-Induced Neurite Outgrowth in Pc12 Cells
Purpose: The present study was conducted to evaluate the effect of different concentrations of morphine on staurosporine-induced neurite outgrowth in PC12 cells. Materials and Methods: PC12 cells were cultured in RPMI1640 culture medium supplemented with 0.2% BSA. Cells were divided into three groups; Ι, ΙΙ and ΙΙΙ, culture in the presence of 50, 100 and 214 nM staurosporine respectively. In ea...
متن کاملModulation of H2O2- Induced Neurite Outgrowth Impairment and Apoptosis in PC12 Cells by a 1,2,4-Triazine Derivative
Introduction: Increased oxidative stress is widely accepted to be a factor in the development and progression of Alzheimer’s disease. Triazine derivatives possess a wide range of pharmacological activities including anti-oxidative and anti-in.ammatory actions. In this study, we aimed to investigate the possible protective effect of 3-thioethyl-5,6-dimethoxyphenyl-1,2,4-triazine (TEDMT) on H2O2-...
متن کاملVinculin-deficient PC12 cell lines extend unstable lamellipodia and filopodia and have a reduced rate of neurite outgrowth
We have studied the role of vinculin in regulating integrin-dependent neurite outgrowth in PC12 cells, a neuronal cell line. Vinculin is a cytoskeletal protein believed to mediate interactions between integrins and the actin cytoskeleton. In differentiated PC12 cells, the cell body, the neurite, and the growth cone contain vinculin. Within the growth cone, both the proximal region of "consolida...
متن کاملSelenium nanoparticles inclusion into chitosan hydrogels act as a chemical inducer for differentiation of PC12 cells into neuronal cells
Background and Objective: Biomaterials and nanomaterials have generated a great opportunity in regenerative medicine. Neurological disorders can result in permanent and severe derangement in motor and sensory functions. This study was conducted to examine the effects of selenium nanoparticles (Se NPs) as a chemical inducer for differentiation of PC12 cells into sympathetic-like neurons characte...
متن کاملNerve growth factor-induced changes in the intracellular localization of the protein kinase C substrate B-50 in pheochromocytoma PC12 cells
High levels of the neuron-specific protein kinase C substrate, B-50 (= GAP43), are present in neurites and growth cones during neuronal development and regeneration. This suggests a hitherto nonelucidated role of this protein in neurite outgrowth. Comparable high levels of B-50 arise in the pheochromocytoma PC12 cell line during neurite formation. To get insight in the putative growth-associate...
